Comments: H36.12d [Pixie 12d]細(xì)胞This cell line was established by P.A. Campbell and E.P. Canono in 1991. Percoll gradient purified, proteose peptone elicited peritoneal macrophage cells from C57BL/6N mice were fused with drug selected P388D1 mouse macrophage tumor cells. [39923]
Unlike H36.12j (ATCC CRL-2449), the cells do not produce tumor necrosis factor alpha (TNF alpha) upon direct beryllium stimulation.
Propagation: ATCC complete growth medium: The base medium for this cell line is ATCC-formulated Dulbecco's Modified Eagle's Medium, Catalog No. 30-2002. To make the complete growth medium, add the following components to the base medium:
heat-inactivated iron supplemented bovine calf serum to a final concentration of 10%
Temperature: 37.0°C
Subculturing: H36.12d [Pixie 12d]細(xì)胞Subcultivation Ratio: A subcultivation ratio of 1:5 every 3 - 4 day is recommended
Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density)
Subcultures are prepared by scraping. Dislodge cells from the flask substrate with a cell scraper. Aspirate gently and dispense into new flasks. Seed flasks at 2 X 10exp5 viable cells/ml.
Preservation: culture medium 95%; DMSO, 5%
